We found that medulloblastomas that form in Smo mutant mice grow more slowly when the animals are genetically engineered so that the gene Bcl2l1 is conditionally deleted in the tumor cells. We undertook this study to define changes in cellular heterogeneity and gene expression patterns in the Bcl2l1-deleted medulloblastomas, compared to medulloblastomas with an intact Bcl2l1 allele. We generated replicate mice that developed medulloblastoma by interbreeding Math1-Cre, SmoM2 and Bcl2l1 f/f mouse lines to generate Math1-Cre/SmoM2/Bcl2l1 f/f and Math1-Cre/SmoM2/Bcl2l1 f/+ mouse pups. We harvested tumors from replicate mice of each genotype at either P12 or P20. We then subjected the tumors to Drop-seq analysis using our previously published methods
