To characterize the site-specific methylation landscape of the Mandarin fish ranavirus (MRV) genome, whole-genome bisulfite sequencing (WGBS) was conducted on an isolated MRV strain. After inoculating Mandarin fish fry (MFF-1) cells with MRV (MG941005.3) at 27°C and 5% carbon dioxide in Dulbecco's modified Eagle medium (DMEM) for 3 days, the sample was collected after repeated freeze-thaw cycles. Virions were purified using a modified double sucrose gradient ultracentrifugation method. Genomic DNA was extracted from the purified virions using the FastPure Cell/Tissue DNA Isolation Mini Kit (Vazyme, China), and whole-genome bisulfite sequencing (WGBS) was subsequently performed.
