To formally assess how 5mC shapes the H3K27me3 landscape, we profiled the epigenome of naive and differentiated cells in the presence and absence of the DNA methylation machinery. Overall design: Whole genome bisulphite sequencing and H3K27me3 and H3K36me3 CUT&Tag was performed on mouse embryonic stem cells and epiblast-like cells. H3K27me3 CUT&Run was performed on epiblast-like cells treated by TET1 site-directed epigenome editing.
