Yeast replication origin with BrdU and IdU

BC01. Yeast with only {T, C, G, A}. BC02. Yeast with BrdU incorporation only. BC03. Yeast with IdU incorporation only. BC04. Yeast with BrdU and IdU incorporation, as you proposed originally. 4 different yeast cultures: without any nucleotide analog (-), with BrdU (Br), with CldU (Cl), and with BrdU and CldU (Br+Cl). Cultures were arrested in G1 phase and released into S phase in the presence of the nucleotide analog for 30 minutes before harvesting. In the case of the Br+Cl culture, cells were released into S phase in the presence of BrdU for 20 minutes, washed with water, and incubated with CldU for 10 additional minutes before harvesting.
