RNA-seq_CCA1-ox_WT

Arabidopsis thaliana WT (Col-0) and CIRCADIAN CLOCK ASSOCIATED 1 (CCA1) over-expressing seeds were surface-sterilized using 15% bleach for 10 min followed by 3 washes with sterilized distilled water. Seeds were then sown on Petri dishes containing Murashige and Skoog (MS) medium containing vitamins (Duchefa), without sucrose. Seeds were stratified at 4ºC in the dark for 4 days, and germination was induced with 6 hours of white light (80 μE) at 22 ºC followed by 18 hours of darkness. Seedlings were then grown under constant light conditions without prior entrainment (15 µE of white fluorescent light) at 22°C and samples were collected at 8 days after germination. Total RNA was isolated using a RNeasy Plant Mini Kit including on-column DNase digestion with the RNase-Free DNase Set (Qiagen). RNA purity was checked using a NanoPhotometer® spectrophotometer (IMPLEN, CA, USA), and RNA integrity and quantitation were assessed using the RNA Nano 6000 Assay Kit of the Bioanalyzer 2100 system (Agilent Technologies).
