Protective Role of Bre1 in Mitochondrial Function and Energy Metabolism in Drosophila Models of Parkinson’s disease

Bre1 is considered to play a crucial role in the clearance of damaged mitochondria, and the impairment of this clearance process is a major contributor to the progression of Parkinson's disease (PD). Therefore, we hypothesize that Bre1 may have a significant role in the development of PD. To test this hypothesis, we generated Drosophila lines with overexpression and knockdown of Bre1 and crossed them with PD model flies. This resulted in PD model flies with either overexpressed or knocked down Bre1.We used RNA-Seq to analyze the gene expression profiles of these flies and found that overexpression of Bre1 led to a significant enrichment of genes related to energy metabolism. Conversely, knockdown of Bre1 increased the expression of many peroxisomal genes. Based on these findings, we propose that Bre1 may improve PD symptoms by enhancing energy supply in PD.
