To better understand the mechanism by which CFZ impacts polyQ toxicity, we conducted a transcriptomic analysis in polyQ94-EGFP expressing U2OS (5µM, 8 days). Data from a transcriptomics experiment in polyQ94-U2OS cells treated with CFZ (5 µM, 8day) was compared to a DMSO-treated control. Overall design: Transcriptomic analysis in polyQ94-EGFP expressing U2OS cells exposed or not to CFZ (5µM, 8days) was performed. Total RNA was extracted from cell pellets using the Agilent Absolutely RNA Miniprep Kit following the manufacturer's instructions. The sequencing library was constructed with the QuantSeq 3' mRNA-Seq Library Prep Kit (Lexogen), and approximately 10 million reads were obtained per sample by Illumina sequencing. STAR was used for sequence alignment based on the GRCh38 DNA primary assembly reference build, and quantification was done using featureCounts(Liao et al., 2014) with reference build GRCh38.101. Differential expression (DE) analyses between the groups were performed using DESeq2.
