Generation Sequencing Facilitates Quantitative Analysis of Wild Type and TaD27 Transcriptomes

To further dissect the molecular basis of tiller generation, we performed an RNA-seq analysis of TaD27-RNAi axillary bud primordia at the three-week old. The axillary buds of wild type KN199 at the same development stage were used as the control. A total of 4,947 differentially expressed genes (DEGs) between the WT and TaD27-RNAi were identified using a fold-change of 2 and FDR<0.05. Among them, 4,124 genes were expressed at a low level, 823 genes were highly expressed in TaD27-RNAi compared to the WT. Wheat mRNA profiles of 3-week old wild type (WT) and TaD27-RNAi were generated by deep sequencing, in triplicate, using Gene Denovo.
