RNAseq was used to identify host and viral transcriptome changes in methionine restricted Rael cells or Rael cells expressing control, AHCY or MAT2A sgRNAs. Overall design: For methionine restriction, Rael cells were washed with PBS and then cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.01mM L-methionine for 3 days. Control cells were cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.1mM L-methionine for 3 days. Control or methionine restricted cells were then subjected to RNAseq analysis. For CRISPR KO, Rael cells expressing control, AHCY, or MAT2A sgRNAs were subjected to the RNAseq analysis.
