Expression of BK channel subunits in HBE and U87-MG cells under senescence-inducing conditions

The experiment was performed on HBE WT and HBE ΔαBK cells, where control cells were not treated and compared to cells treated with 150 µM H₂O₂ or 50 nM doxorubicin, as well as on U87-MG WT and ΔαBK cells, where control cells were not treated and compared to cells treated with 300 µM H₂O₂ or 20 nM doxorubicin. Gene expression was analyzed by RT- PCR using primers specific for genes encoding different isoforms of the BK channel α subunit, including DEC, STREX, VYR, and the glioma-specific isoform gBK. GAPDH and TBP were used as reference genes.Under the same experimental conditions, PCR reactions were performed to determine the expression levels of the β and γ subunits of the BK channel.
