We have developed a high-throughput genetic screen combining flow cytometry sorting with next-generation DNA sequencing of enriched genomic libraries, which has revealed known and novel mediators of reversible filamentation in uropathogenic E. coli (UPEC). Confirmation of a subset of genes and partial genomic regions verified our results. This approach offers the opportunity to carry out comprehensive screens of diverse DNA libraries in order to identify new DNA sequences encoding the capacity to inhibit bacterial cell division, or other selectable traits.
