SOLiD sequencing of clonally expanded murine intestinal stem cells

Single GFP-positive adult stem cells were isolated from the small intestine of a Lgr5 reporter mouse. These cells were clonally expanded in 3D organoid culture to obtain sufficient DNA for whole genome sequencing analysis. After 56 days of in vitro expansion from each original cloanl culture a second clonal step was performed (subclone), followed by a new round of WGS analysis. Sequening was performed by SOLiD sequencing to an average base coverage of ~ 18x.
