Cryptic splicing in massively parallel reporter assays

The goal is to assess the prevalence and consequence of cryptic splicing in massively parallel reporter assay (MPRA). We used PTRE-seq as a representative MPRA experiment. The PTRE-seq library was transfected into four different cell types, followed by RNA isolated and sequencing. Spliced fraction was assessed using a custom computational pipeline for each reporter in the library.
