The Citrobacter TSA-1 bacteria is found in the hindgut of termites. Our project will aid in the search for the gene that is responsible for pentavalent arsenic(As(V)) respiration. No homolog for respiratory arsenate reductase(arrAB) nor amplicons of arrA were found using the previous genomic assembly. In order to achieve the best assembly, we separated the process into three major milestones. The first step was to extract purified high molecular weight DNA, then to prepare and sequence the genome with the Oxford Nanopore Technologies MinION, and to finally run the genome through an assembly pipeline engineered by us. The final assembly will be a polished hybrid assembly that uses both illumina short reads and MinION long reads.
