Seawater 16S amplicon sequencing

Here, we report a developed process that the enriched microbial community can simultaneously synthesize C1-C6 alkanes and C2-C3 alkenes with using CO2 as solo carbon source under photosynthetic condition, which is achieved in a distinguished way from previous studies on the production of short chain hydrocarbons, including two sections, namely the section of carbon fixation and hydrocarbon synthesis. Seawater samples were fetched from beach and filtrated for cell collection. Total DNA was isolated from 30 ml of each sample using the QIAamp DNA Mini Kit (Qiagen, California, USA) according to the manufacturer's protocol. The V3-V4 region of 16s rDNA was amplified using primer sets Bac336F/Bac806R for bacteria and Arc334F/Arc806R for Archaea, which were subjected to Illumina sequencing at the Miseq platform.
