FKHD-mutant (FKHDm) FOXA1 has impaired ability to bind chromatin and thus failed to rescue wildtype FOXA1-mediated transcriptional program. FKHDm FOXA1 formed much stronger interaction with the AR protein, thereby preventing AR from binding to the chromatin. Microarray analysis on LNCaP shCtr and shFOXA1 cells with overexpressed FOXA1 wildtype or various FOXA1 mutants in the presence or absence of androgen (R1881).
