Rec8 is a meiosis-specific subunit of the cohesin complex in Saccharomyces cerevisiae and plays a critical role in sister chromatid cohesion (SCC). Previous studies have identified two chromatin-bound pools of cohesin: a stable pool essential for SCC and a dynamic pool whose functional significance remains elusive. In this study we used the anchor away technique for the conditional depletion of dynamic pool of Rec8-cohesin using Rapamycin (by fusing REC8 with FRB-GFP). To quantify changes in chromatin-bound Rec8 upon rapamycin-induction, we performed ChIP-seq in the S. cerevisiae SK1 background. We used anti-mTOR antibodies (Enzo Life Sciences, ALX-215-065-1) to pull down Rec8-FRB-GFP. The submitted files contain raw sequencing data for Rec8 ChIP-seq, generated to assess Rec8 occupancy and distribution under control and rapamycin-treated conditions.
