Cellular sources of liver endothelial cells remain elusive. Here, we used irradiation-conditioned bone marrow chimeric mice to lineage trace the endothelial cells. Bone marrow-derived (YFP+) and resident (YFP-) liver endothelial cells were isolated and total RNA were used for microarray analysis using Clariom S mouse assays (Applied Biosystems). We generated bone marrow chimeras by transplanted 5000 YFP-labeled LSK cells in wild-type C57BL\6N mice which were irradiated with 9 Gy (2 x 4.5 Gy) radiation. After one month, the successful bone marrow chimeric mice were sacrificed and liver endothelial cells were isolated by FACS. Further, liver EC were segregated into YFP+ (bone marrow-derived endothelial cells) and YFP- (resident liver endothelial cell). Total RNA was isolated with Arcturus PicoPure RNA isolation kit. cDNA libraries were prepared and analyzed by the DKFZ Genomics core facility, according to the standard operating protocols.
