Exome capture sequencing of 96 winter and spring barley cultivars for the analysis of genome-wide haplotype diversity

We exome captured and sequenced 96 elite two-row barley (Hordeum vulgare) cultivars in order to carry out a genome-wide analysis of haplotype patterns. This included 48 winter lines and 48 spring lines. The Roche Nimblegen exome capture array was used to enrich for exonic sequence (http://sequencing.roche.com/products/nimblegen-seqcap-target-enrichment/seqcap-ez-system/seqcap-ez-designs.html). The enriched samples were then sequenced on an Illumina HiSeq 2500 sequencer. An average of 2 x 21,876,780 paired end Illumina reads per sample was generated. This equates to approximately 72x coverage of the 61 Mbp exome capture space.
