This study investigates the transcriptional effects of TSA and Aß oligomers on human neuronal cells using RNA sequencing of iPSC-derived cortical neurons subjected to various treatment conditions. The experimental design included four distinct groups: untreated control, TSA treatment alone, Aß exposure alone, and combined TSA treatment and Aß exposure. Each condition was evaluated with four biological replicates, with an additional technical replicate for each condition to assess reproducibility. Overall design: RNA-seq analysis of iPSC-derived cortical neurons with 4 treatment conditions: (1) Control (untreated), (2) TSA treatment alone (60 ng/ml), (3) Aß oligomer exposure alone (5 µM), and (4) Combined TSA pre-treatment (60 ng/ml for 24h) followed by Aß oligomer exposure (5 µM). Cells were treated at DIV 4 and RNA was extracted using Total RNA Purification Kit. Four biological replicates per condition with additional technical replicates.
