The DNA cleavage efficiency of the exoCasMINI system.[Deep-seq]

The fusion of a T5 exonuclease to the C-terminus of CasMINI (exoCasMINI) could indeed enhance the DNA cleavage efficiency in mammalian cells. Overall design: To investigate the potential of exoCasMINI in genomic DNA cleavage, we transfected CasMINI or exoCasMINI or SpCas9 or LbCas12a into MCF7 cells and HEK292T cells with gRNA targeting different genomic sites. The results indicated that exoCasMINI significantly improved editing efficiency at all the tested sites compared to CasMINI.When compared to SpCas9 and LbCas12a, exoCasMINI displayedsimilar DNA cleavage activity at 5 of tested sites,while the other loci remain lower activity.
