To generate iPGC1α-/- mice, PGC1αfl/fl mice were intercrossed with Vil1-Cre mice to obatin Vil1-CreTg/-PGC1αfl/- mice. These mice were then backcrossed with PGC1αfl/fl mice to restore homozygosity for the iPGC1α floxed allele. Male Vil1-CreTg/-PGC1αfl/fl mice were bred to PGC1αfl/fl to produce the mice used in the study. PGC1αfl/fl (iPGC1afl/fl) littermates were used as controls. Eight-weeks old mice were treated for 8 weeks weeks with 42% Kcal/fat diet enriched with saturated fats (0.2% total cholesterol, milk fat) (Teklad, TD.180342). Chow diet was used as control. Mice were randomly assigned to treatment groups for in vivo studies. All mice were sacrificed randomly fasted at ZT 3. For ileum section, the isolation of single cells was performed by a modification of Weiser method.
