RNA-seq of maize root hair tissue in response to mild and severe cold stress

Seeds of the maize genotype 'B73' were grown in paper rolls soaked with ddH2O for 3 days under control conditions. 3 days after germination in paper rolls under control conditions (27 C/22 C for 16 h light/8 h dark), seedlings were subsequently subjected to control, mild cold (16 C /12 C for 16 h light/8 h dark) or severe cold (10 C /6 C for 16 h light/8 h dark) treatment without preselection. Sampling was carried out immediately after each treatment. Roots were shock frozen in liquid nitrogen, and root hairs were dislodged and collected. Root hair tissue for each individual sample was pooled from 120 seedlings per sample. RNA was isolated from individual samples and sent for library preparation and sequencing.
