miRNA-based liquid biopsy provides early prediction of lethal acetaminophene poisoning: a case study

The study comprises of 24 samples that have been taken over a time course during a lethal APAP overdose Samples have been taken after an accidental acetaminophen overdose from a single case. Several blood samples were taken per day during the treatment until the subject deceased. Samples were recovered from whole blood following a centrifugation of whole blood at 3000g for 10 minutes at room temperature and subsequently stored at 80°C. Circulating miRNAs were isolated using the miRNeasy Serum/Plasma kit (Qiagen) from 50ul serum; the quality of the RNA was evaluated using a Bioanalyzer 2100 using the small RNA Kit (Agilent). Sequencing libraries were prepared using the TruSeq Small RNA-Seq Preparation Kit (Illumina), and sequenced on a HiSeq 2000 (Illumina) according to the manufacture's protocol.
