UVB plays a key role in inflammation and DNA damage in human skin. Human keratinocyte HaCaT cells were utilized to determine the up- and down-regulated miRNA after UVB exposure. We used microarrays to identify the miRNA affected after UVB (15 mJ/cm^2) exposure to HaCaT cells Overall design: HaCaT cells were exposed to UVB (15 mJ/cm^2) and incubated for 6 hours. The cells were collected usign TRI reagent, RNA was extracted, and subjected to Affymetrix microarrays. We sought to investigate miRNA regulating UVB-induced inflammation.

紫外线B(UVB)在人类皮肤的炎症反应与DNA损伤中发挥关键调控作用。本研究以人角质形成细胞HaCaT系为模型,探究UVB暴露后表达上调与下调的微小RNA(microRNA, miRNA),并通过微阵列技术鉴定15 mJ/cm²剂量UVB照射HaCaT细胞后受影响的miRNA。实验设计如下:将HaCaT细胞经15 mJ/cm² UVB照射后孵育6小时,使用TRI试剂收集细胞,提取总RNA后进行Affymetrix微阵列检测。本研究旨在探究调控UVB诱导皮肤炎症的miRNA。
