Skin cells were depleted from CD45+ and dead cells using microBeads, purity 95%. CD45- cells were RNA sequenced (bulk) to find differentially regulated genes between healthy donors and scleroderma patients (SSc) Overall design: CD45- cells were isolated from 3 punch biopsies (4mm) od 3 patients or healthy donors. Samples were group-pooled to compensate for HLA-DR differences.
