IEAtlas reanalyzed publicly available mass spectrometry-based HLA immunopeptidome datasets against our integrated benchmarked non-canonical open reading frame information. The current IEAtlas identified 245 870 non-canonical epitopes binding to HLA-I/II allotypes across 15 cancer types and 30 non-cancerous tissues, greatly expanding the cancer immunopeptidome. IEAtlas further evaluates the immunogenicity via several commonly used immunogenic features, including HLA binding affinity, stability and T-cell receptor recognition. In addition, IEAtlas provides the biochemical properties of epitopes as well as the clinical relevance of corresponding genes across major cancer types and normal tissues.
IEAtlas以我们经整合并经基准验证的非经典开放阅读框(open reading frame,ORF)信息为参照,重新分析了公开可得的基于质谱(mass spectrometry)的人类白细胞抗原(Human Leukocyte Antigen,HLA)免疫肽组数据集。当前版本的IEAtlas已在15种癌症类型与30种非癌组织中,鉴定出245870个可与HLA-I/II异型结合的非经典表位,极大扩充了癌症免疫肽组库。IEAtlas还通过多项常用免疫原性特征对免疫原性进行评估,涵盖HLA结合亲和力、稳定性以及T细胞受体识别特性。此外,IEAtlas还提供了表位的生化属性,以及对应基因在主要癌症类型与正常组织中的临床相关性。
