Next Generation Sequencing Analysis of OX40hi Treg and OX40lo Treg cells from mouse tumors and spleens

Based on RNA-seq technology, we studies gene expression profiles in Treg cells isolated from mouse tumors and spleens. We identified differential gene expressions comparing OX40hi Treg with OX40lo Treg cells from tumors and spleens. The difference of Trges in tumors and spleens were also identified. Foxp3-GFP mice were intradermally implanted with B16 cells at 5x10^5 cells at the right side. After 7-10 days, tumors and spleens were dissected and digested into single cells. After staining with antibodies against CD45, CD3, CD4 and OX40, OX40hiGFP+ and OX40loGFP+ cells were sorted in BD FACSAria cell sorter.
