The outer retina in young P23H retinitis pigmentosa mice overexpresses critical components of synaptic transmission

To determine the gene expression changes in the retina of 1-month-old P23H/Gnat2-KO versus Gnat2-KO mice, we generated single-cell suspensions, and performed scRNA-Seq (10X Genomics). Overall design: One retina from two male and two female mice per group (total n=4 per group; groups: Gnat2-KO and P23H/Gnat2-KO mice) were pooled to prepare two single-cell suspensions at a final concentration of 1,000 cells/µl. Freshly dissociated cells (~10,000) were loaded into a 10x Genomics Chromium Single Cell system using v3 chemistry following the manufacturer's instructions.
