Comparative transcriptomic analysis of max2, hxk1 mutant and Wild type seedlings treated with glucose in early seedling development

A. thaliana seeds of max2, hxk1, and WT were surface-sterilized and allowed to imbibe water in the dark at 4 ?C for 3 d. The seeds were then transferred to Petri dishes containing water with 2% glucose or mannitol. The cultures were shaken at 140 rpm at 23 ?C for 3d under fluorescent white light with a 16 h light/8 h dark photoperiod.The seedlings were collected and rinsed three times with sterile distilled water, and excess water was removed by centrifugation. Total RNA was extracted from the seedlings and conducted a comparative transcriptomic analysis using the short-read sequencing technology (Illumina) combined with a tag-based digital gene expression (DGE) system.
