How sleep influences the transcriptional programing of blood leukocytes in humans remains incompletely undertstood. Volunteers with adequate sleep participated in a 6 week cross-over design trial. During the habitual sleep (HS) phase, participants did not alter their sleep and slept adequately. During the sleep restriction phase (SR), participants restricted their sleep by ~1.5 hours per night. PBMCs from 8 individuals (4 HS and 4 SR) were collected subjected to scRNAseq. Overall design: Peripheral blood mononuclear cells (PBMCs) were collected in the evening (target time: 15:00â17:00) from men and women, aged 20â75 y with BMI <35 kg/m2, participating in a 6-week randomized crossover study in which chronic mild insufficient sleep was imposed on volunteers with adequate habitual sleep duration. To examine if sleep shapes the transcriptional landscape of monocytes, we performed scRNAseq of PBMCs on eight distinct participants whose sleep duration, measured objectively with nightly actigraphy, was 7.5±0.1 hours/night during the habitual sleep (HS) phase and 6.2±0.1 hours/night during the sleep restriction (SR) phase.
