Single-cell measurements and MATLAB analysis code supporting Ginzberg, Tan, Patel and Kafri, "Inferring feedback regulation from static snapshots of a single signal" (Biochemistry and Biophysics Reports). AoF (Assay of Feedback) combines fixed-cell single-cell imaging with ergodic rate analysis to infer the feedback spectrum phi(l) of a signal across the cell cycle, without pathway perturbation or prior knowledge of circuit topology. Applied to pAkt(Ser473) in HeLa cells, it reveals a negative-feedback trough near the G1/S transition (phi = -1.70 at 24% of cell-cycle progression, 95% BCa CI [-2.02, -1.39], BH-FDR q = 1.8e-23) that is abolished by rapamycin. This deposit contains the two single-cell feature tables (489,798 control and 190,249 rapamycin-treated cells; columns DNA, logGeminin, pAkt, and cell-cycle position l), the 402-node cell-cycle trajectory that defines l, the ergodic-rate-analysis estimator, the bootstrap and false-discovery-rate analysis with its precomputed output, and the script that renders Figure 1 panels A to D. They are wet-lab measurements, are not reproducible from code, and are not included here.
