In order to explore and compare the transcriptomic profile of hypodermal adventitia fibroblasts, fibroblasts were isolated via flow cytometry mouse hypodermis and were subjected to RNA-sequencing analysis. Overall design: Back skins from age-matched Csf1f/f and Tek-crexCsf1f/f mice were harvested and hypodermal adventitial was isolated by mechanical force and enzymatic treatments. Cells were liberated after enzymatic digestion of the tissues. Fibroblasts (CD45- CD31- Sca-1+ CD34+) were isolated by fluorescence-activated cell sorting (FACS) and their transcriptome assessed by RNA-sequencing.
