Differential gene expression analysis was performed to assess the affects of the deletion of sRNA SR0726 on the in vitro transcriptome of Borrelia burgdorferi in order to investigate a potential regulatory role for the sRNA Triplicate cultures of wild type and D7 (SR0726 deletion mutant) were grown in BSK-II under standard conditions until mid-log phase, sub-cultured (1:100) to room temperature BSK-II, grown back to mid-log phase, subcultured (1:100) to 35°C, and grown to late-phase. Triplicate cultures for each strain were then pooled and pelleted. RNA was extracted, DNase treated, and was sent to the Novogene Corporation for library preparation, sequencing, and bioinformatic analysis.
