HEK293T cells were transfected with the plasmid construct expressing FLAG-GST-tagged SUGP1 (aa 543-645), followed by two rounds of affinity purification using FLAG and GST tags. The purified proteins were resolved by SDS-PAGE, followed by staining with QC Colloidal Coomassie Stain (Bio-Rad). The relevant gel region (~85-100 KD) was excised, and the proteins in this gel section were identified by mass spectrometry.
