We engineered a H1 knockout mutant in Arabidopsis thaliana which shows pleiotropic phenotypes. Using High-throughput Chromosome Conformation Capture (Hi-C), we found that H1 depletion dampens inter- and intra-chromosomal interactions.our results provided insights into the roles of linker histone H1 in mediating chromatin conformation and coordinating DNA methylation and gene expression.
