Gene expression changes in A375 melanoma cells in response to ERK3 knockdown.

Gene expression microarrays were used to compare A375 cells treated with a control siRNA or an siRNA targeting ERK3 (MAPK6). Three replicates are included for each condition (siCon and siERK3). Total RNA was extracted using Trizol® reagent (Ambion) three days post-siRNA-transfection. RNA quality was assessed on an Agilent 2100 Bioanalyzer following the manufacturer’s protocol. Samples with RNA integrity numbers (RIN) >6 were used for further analysis. RNA concentration was measured by NanoDrop™ One Spectrophotometer (Thermo Fisher Scientific). Affymetrix microarray system was used for gene expression analysis using Clariom™ S Pico Assay (human) platform with WT PLUS Kit and GeneChip® Hybridization, Wash, and Stain Kit following the manufacturer’s protocols.
