ER, mitochondria and ISR regulation by mt-HSP70 and ATF5 upon procollagen misfolding (E18.5 femur and tibia)

We used single cell RNA sequencing (scRNA-seq) to identify femur and tibia osteoblast cell stress response pathway to misfolding of type I procollagen with Gly610->Cys substitution in the triple helical region of alpha2(I) chain. Femur and tibia cells were isolated from E18.5 embryos as described in Gorrell et al, Adv Sci, 2022本研究采用单细胞RNA测序（single cell RNA sequencing, scRNA-seq），旨在鉴定α2(I)链三螺旋区域中存在Gly610→Cys取代的I型前胶原错误折叠所对应的股骨与胫骨成骨细胞应激反应通路。我们参照Gorrell等人2022年发表于《Advanced Science》的实验方法，从胚胎发育第18.5天的胚胎中分离得到股骨与胫骨细胞。
