By introducing point mutations in the RNAse III domains of plant DCL1 we generated Catalytically inactive (ci) DCL1. Plant expressing DCL1ci show an increase stability of the RNA/DICER complexes and therefore represent an innovative strategy to perform a direct and systematic analysis of DCL1 substrates.
