Single-cell and bulk RNA sequencing of PDX tumors or cell models before and after drug treatment

Subcutaneous PDX tumors (PDX #73, #91, #419) were established in female B-NDG mice using patient-derived HCC tissues. Tumors were randomized for FDI-6 (30 mg/kg, i.p., every other day) or control treatment. Tumor samples were collected at day 16 for 10x single-cell RNA-seq and bulk RNA-seq. Bulk RNA-seq was performed for Huh7 (15 μM, 4 and 7 days) and HepG2 (20 μM, 2 and 4 days) cells following FDI-6 treatment. For Huh7 cells, additional groups were treated with IFNγ (10 ng/mL) alone or in combination with FDI-6 (15 μM) for 4 days prior to bulk RNA-seq.
