<em>Plectropomus leopardus</em>, as known as leopard coral grouper, is a valuable marine fish that has been gradually artificial breeding. To promote future conservation, molecular breeding and comparative studies, we generated an improved high-quality chromosomal-level genome assembly of leopard coral grouper using Nanopore long-reads, BGI short reads, and the Hi-C sequencing data, as well as protein-coding genes predicted <em>de novo</em> and verified using sufficient transcriptome data. The draft genome of 849.74Mb with 45 contigs and N50 of 35.59 Mb. Finally, a total of 846.49 Mb (99.6%) of based were anchored to 24 pseudo-chromosomes using Hi-C technology. A final set of 27,285 genes is annotated after manual curation of the predicted gene models, and BUSCO analysis yielded a completeness score of 99.5%. This study significantly improves the utility of the grouper genome and is a reference for the study of molecular breeding and fish genomics and biology.
豹纹鳃棘鲈(Plectropomus leopardus),俗称豹纹珊瑚斑(leopard coral grouper),是一种具有较高经济价值的海水鱼类,目前已逐步实现人工繁育。为推动后续的物种保护、分子育种及比较研究,本研究利用纳米孔(Nanopore)长读长测序数据、BGI短读长测序数据及Hi-C测序技术,并结合从头(de novo)预测的蛋白编码基因与充足的转录组数据,完成了该物种的改进型高质量染色体级基因组组装。本次组装得到的草图基因组大小为849.74 Mb,包含45个重叠群(contig),重叠群N50值为35.59 Mb。随后通过Hi-C技术,将其中99.6%(共计846.49 Mb)的序列锚定至24条伪染色体上。经对预测基因模型进行人工校正注释后,最终共得到27285个蛋白编码基因;BUSCO(Benchmarking Universal Single-Copy Orthologs)分析结果显示,基因组完整度得分为99.5%。本研究显著提升了石斑鱼基因组的应用价值,可为分子育种、鱼类基因组学及鱼类生物学研究提供重要参考依据。
