We identified a tumor suppressor gene Ptp61F, which could synergize oncogenic gene Ras to drive tumorigenesis in Drosophila epithelial tissue, while Ptp61F homozygous mutant larvae showed defects during development. In order to dissect the underlying molecular mechanisms and crosstalks of different signaling pathways, we applied RNA-seq in imaginal eye discs as well as Ptp61F homozygous mutant larvae. Here, we found Hippo signaling, Tor signaling, and endocytosis pathway were dramatically altered. And the following experiments proved their functions in regulating Ptp61F mediated three-dimensional (3D) organ size. Our findings provide insights into how 3D organ size is controlled under both physiological and pathological conditions.
