We generated single nucleus transcriptomic atlas for white and brown adipose tissue from adipocyte-specific Gls knockout mouse, and also generated bulk RNA-seq from in vitro differentiated human white adipocytes with/without GLS knock down. Overall design: For snRNA-seq, brown and white adipose tissues were isolated from adipocyte-specific Gls knockout mice and Cre- neagtive Littermates and flash frozen in liquid nitrogen. The nuclei was extracted and performed snRNA-seq. For bulk RNA-seq, in vitro differentiated white adipocytes where transfected eight days post beginning of differentiation, with scrumble or GLS targeting siRNA, cells were harvested and total RNA extracted at D13 of differentiation.
