we have successfully established an in vitro iPSC-derived vascular organoid (iVO) model that can reproduce the vascular injury and inflammatory phenotype associated with the viral infection. In particular, we identified a marked increase in the expression of a part of the membrane attack complex, an intermediate and final product of the complement pathway, and molecules that catalyze upstream reactions (classical; C1QA, C1QB, C1QC, and alternative; CFD) We developed an infection-competent human vascular organoid from pluripotent stem cells amenable for modeling endotheliopathy and performed transcriptome analysis at 4 days-post-infection of SARS-CoV-2 to probe infected endothelial cell-specific signatures.
