To evaluate the differential effects of acetate on microbe-monocolonized mice, B220+IgA+ cells in colonic lamina propria was collected from C57BL/6 mice monocolonized with Bacteroides thetaiotaomicron or Escherichia coli. RNA was extracted and purified using Agencourt RNAClean XP (Beckman Coulter). cDNA was synthesized via reverse transcription-PCR using the primers specific for constant region of heavy chain alpha. Sequencing was performed on MiSeq platform (Illumina, 400 bp for read1 and 100 bp for read2).
