Developmental trajectory of pre-hematopoietic stem cell formation from endothelium (scRNA-seq data set)

To examine the cell and molecular transitions between endothelial (E), hemogenic endothelial (HE), and intra-arterial clusters (IAC) cells, and the heterogeneity of hematopoietic stem and progenitor cells (HSPCs) within the IACs, we profiled ~40,000 cells from the caudal arteries (dorsal aorta, umbilical, vitelline arteries) of embryonic day (E) 9.5 to 11.5 mouse embryos by single-cell RNA sequencing (scRNA-Seq). We identified a developmental bottleneck in endothelial to hematopoietic transition (EHT) and heterogeneity within the IAC population. Single cell RNA-Seq profiles of cells involved in endothelial to hematopoietic transition (EHT) from mouse embryos at embryonic day 9.5, 10.5, 11.5 and functional HSCs at E14.5.
为探究内皮细胞(endothelial, E)、生血内皮细胞(hemogenic endothelial, HE)与动脉内簇(intra-arterial clusters, IAC)细胞之间的细胞及分子转化过程,以及动脉内簇中造血干细胞和祖细胞(hematopoietic stem and progenitor cells, HSPCs)的异质性,我们对胚胎日(embryonic day, E)9.5至11.5的小鼠胚胎的尾动脉(包含背主动脉、脐动脉与卵黄动脉)中的约40000个细胞开展了单细胞RNA测序(single-cell RNA sequencing, scRNA-Seq)分析。本研究鉴定出内皮向造血转化(endothelial to hematopoietic transition, EHT)过程中的发育瓶颈,以及动脉内簇群体内的异质性。本数据集同时涵盖胚胎日9.5、10.5、11.5小鼠胚胎中参与内皮向造血转化过程的细胞的单细胞RNA测序图谱,以及E14.5时期功能性造血干细胞(hematopoietic stem cells, HSCs)的测序数据。
