ddRAD sequences of Hyperolius substriatus across the East African Range

Hyperolius substriatus (African spotted Reed Frog) is a highland-adapted small frog from East Africa. Distributed throughout the highland areas of Tanzania, Malawi, and Mozambique, subspecies and molecularly-distinct lineages have been identified, but uncertainty in the phylogenetic relationship of these lineages using pre-genomic methods made assessing these potential subspecies difficult. With the addition of ddRAD-seq derived genomic material and additional geographic sampling throughout poorly understood regions, we identified 3 primary lineages: a Tanzanian lineage which contains the type locality for Hyperolius substriatus (East Usambara Mountains), a northern Malawian lineage (Misuku Hills, Nyika Plateau, and Vipya Highlands), and a southern Malawian and central Mozambican lineage (containing isolated mountain blocks including Chiperone, Socone, and Namuli in Mozambique; Mulanje and Cholo in Malawi).
