In the BY4741 background, we constructed two genetic systems allowing to generate a duplication on chromosome XV and a translocation between chromosomes IV and X, respectively. Yeast colonies were grown from single cells on a non-selective synthetic media. Colonies were spread on selective media to select either for the presence of the translocation and the duplication or the duplication and the canavanine resistance. The set of 12 strains includes the parental strain with the genetic systems as well as 2 mutants carrying both the duplication and translocation and 9 mutants carrying both the duplication and mutations in CAN1 gene, isolated in the progeny of the parental strain.
