CARD14 is encoded by a gene expressed predominantly in keratinocytes but also placental tissue, endothelial in the dermis and some immune cells. Mutations in this gene have been found to cause psoriasis and other skin diseases in human patients. Mutation of the E138 residue in the coiled-coil domain of CARD14 has been associated with pustular psoriasis and pityriasis rubra pilaris. Heterozygous mice harbouring a deletion of amino acid E138 in the Card14 gene (generated using CRISPR/Cas9 technology) results in transgenic mice that spontaneously develop a chronic psoriatic skin phenotype particularly on the ear and tail. RNAseq was performed to compare the transcriptome of 8 week-old heterozygous Card14delE138 mice against wild-type littermates.
