Objective Based on previous studies how high salt diet affects cellular immunity,, this study explored how high salt diet affects humoral immunity and its possible molecular mechanisms of action..Methods KLH/CFA mice were randomly divided into a control group and a high salt diet group. The effects of high salt diet on the activity, proliferation, function, and differentiation of T and B cells were examined by flow cytometry; In addition, splenic T and B cells isolated mice were cultured, activated, and induced for the generation of activated B cells under iTreg and LPS stimulation. We further analyzed their mechanism of action under high salt diet.Results ① A high salt diet significantly promoted the Tfh amplification and humoral immunity. ② High salt diet significantly enhanced IL-2 production and cell proliferation in CD4+ T cells. ③ High salt diet markedly increased the plasma cell differentiation and facilitated IL-2 secretion in vitro. To our knowledge, it is first time that a high salt diet regulates humoral immunity by modulating Tfh expression and affecting IL-2. This helps provide new insights into the prognosis and dietary guidance for clinical treatment and humoral immune related diseases.
